Prisys Biotech Establishes Cynomolgus Monkey Psoriasis Model To Accelerate Drug Development

Psoriasis is a multifactorial immune-mediated disease characterized by systemic inflammation and immune dysfunction. It manifests as itchy, scaly plaques that significantly impair the quality of life and mental well-being of patients. These lesions commonly appear on the knees, elbows, trunk, and scalp. Current therapeutic approaches aim to slow excessive skin cell growth and alleviate symptoms. These include topical treatments, phototherapy, and systemic therapies such as immunosuppressants and biologics. Recent guidelines from the American Academy of Dermatology and the National Psoriasis Foundation (AAD/NPF) recommend biologics as a first-line treatment for moderate-to-severe plaque psoriasis, given their high specificity, efficacy, and safety (JAMA, 2020; 323(19):1945–1960). Consequently, biologics have become a focal point in psoriasis drug development.

Development of the Cynomolgus Monkey Psoriasis Model

Prisys Biotech employs the widely recognized imiquimod (IMQ)-induced method to establish a psoriasis model. By administering a precise dose of IMQ to healthy cynomolgus monkeys, we mimic the pathological changes observed in human psoriasis. In our studies, we categorized animals into three groups: a normal control group, an IMQ-induced model group, and a treatment group receiving calcitriol. Following IMQ induction, the model and treatment groups exhibited distinct psoriasis-like lesions. Remarkably, calcitriol treatment in the positive drug group significantly ameliorated these skin abnormalities. Clinical scoring across all groups revealed significant differences, underscoring the success of the model.

Pathological Insights and Clinical Relevance

To further investigate the progression of psoriasis, we collected skin samples at multiple stages: pre-induction, induction, post-induction, treatment, and post-treatment. Histological analyses revealed hallmark psoriasis features in the model and treatment groups, including inflammatory cell infiltration, parakeratosis, and hyperkeratosis. Notably, symptoms in the treatment group improved substantially after calcitriol administration, as evidenced by histopathological findings.
