Product Introduction

Nose-to-Brain (N2B) Drug Delivery: Accelerating CNS Drug Development

Prisys Biotech has developed a dedicated Nose-to-Brain (N2B) drug delivery platform to support the development of central nervous system (CNS) therapeutics. This innovative solution aims to overcome the challenges of the blood-brain barrier (BBB) by enabling efficient and targeted delivery of drugs directly to the brain via the intranasal route.

Our N2B services are designed to facilitate translational studies in non-human primates (NHPs), offering researchers an effective pathway for evaluating drug delivery, distribution, and pharmacodynamics in vivo.

Core Technology and Objectives

The Prisys N2B platform leverages advanced delivery systems, and specialized NHP devices. These devices are customized with NHP-adapted nozzles to optimize dosing precision.

Key optimization parameters include:

  • Nozzle geometry: Adjustable extension lengths and diameters.
  • Material design: Metal nozzles coated with a thin silicone layer to minimize tissue irritation.
  • Formulation compatibility: Especially suited for dry powder formulations, with tracer agents incorporated for verification.
High-density contrast agent strips can be seen in the olfactory mucosa area of the superior nasal meatus (coordinate center)

Imaging Validation and Functional Assessment

Prisys Biotech integrates advanced clinical imaging technologies to confirm and quantify intranasal delivery outcomes:

CT and MRI Validation:

  • Dry powder formulations mixed with contrast agents allow real-time visualization of deposition.
  • CT scans demonstrate concentrated linear contrast signals localized in the olfactory epithelium of the upper nasal cavity.
  • MRI scans using diluted contrast agents provide consistent confirmation of deposition sites.
MR contrast medium hyperintense images can be seen in both superior nasal meatus and olfactory mucosa areas (arrows)

Quantitative Biodistribution and Pharmacodynamic Studies

To further support CNS drug development, Prisys offers extended N2B evaluation services:

  • PET-CT scanning: Using radiolabeled compounds to assess quantitative biodistribution.
  • Histological evaluation: Post-mortem tissue analysis with immunohistochemistry (IHC) or immunofluorescence (IHF) to determine regional distribution of delivered agents.

These integrated approaches enable clients to evaluate both delivery efficiency and pharmacological activity within the CNS.
